Silent decision: HP1 protein escorts heterochromatic RNAs to their destiny

A recent study published in Molecular Cell reports on a heterochromatin-specific checkpoint that captures and primes heterochromatic RNAs for degradation. The authors propose HP1Swi6 to facilitate this by competitive binding to mRNA transcripts respective histone tails.

Impairment of GABAB receptor dimer by endogenous 14-3-3ζ in chronic pain conditions

Complexin arrests a pool of docked vesicles for fast Ca2+-dependent release

Regulated exocytosis of synaptic vesicles is triggered by a local increase in calcium levels, relieving a block in the assembly of the membrane fusion machinery. Based on a reconstituted system, this study characterizes the roles of complexin and synaptotagmin 1 in arresting and synchronizing fusion.

BiP-mediated closing of the Sec61 channel limits Ca2+ leakage from the ER

Sec61, the protein-conducting channel of the endoplasmic reticulum, also mediates calcium efflux. The chaperone BiP binds to a luminal loop of Sec61 to inhibit calcium leakage and to link channel gating to protein homeostasis in the endoplasmic reticulum.

Dephosphorylation of Cdc20 is required for its C-box-dependent activation of the APC/C

The WD40 protein Cdc20 activates the ubiquitin ligase APC/C during mitosis. CDK phosphorylation inhibits Cdc20 binding to APC/C and requires specific reversal by phosphatases, paralleling the control of the related activator Cdh1 in later stages of mitosis.

Altered BCR signalling quality predisposes to autoimmune disease and a pre-diabetic state

The related tyrosine kinases Syk and Zap-70 are key signalling proteins downstream of antigen receptors. A knock-in strategy reveals that the two kinases differentially affect BCR signalling, leading to aberrant B cell section and increased risk of autoimmune disease.
