Downregulation of c-Myc is involved in TLR3-mediated tumor death of neuroblastoma xenografts

This study demonstrates that high c-myc protein expression may increase tumor cell sensitivity to the toll-like receptor 3 (TLR3) agonist-polyinosinic-polycytidylic acid treatment in MYCN-non-ampli?ed neuroblastoma xenografts. Downregulation of c-myc might be involved in TLR3-mediated tumor growth arrest and reactive oxygen species-mediated apoptosis. Therefore, c-myc protein expression may play an important role in TLR3-induced innate immune responses, providing future treatment options.

Alpha-2, 3-sialyltransferases regulate the multidrug resistance of chronic myeloid leukemia through miR-4701-5p targeting ST3GAL1

The expression of the sialyltransferase ST3GAL1 was found to correspond to the drug-resistant phenotype of chronic myeloid leukemia (CML) cell lines both in vitro and in vivo. Micro RNA (miR)-4701-5p directly targets ST3GAL1 to reduce CML cells' resistance. These results indicate that interfering with miR-4701-5p and ST3GAL1 may be of therapeutic value in CML.

ILK–PI3K/AKT pathway participates in cutaneous wound contraction by regulating fibroblast migration and differentiation to myofibroblast

Integrin-linked kinase (ILK) is a key mediator in integrin signaling, which regulates the interactions between fibroblasts and extracellular matrix in wounds. This study found that ILK participates in wound and the fibroblast-populated collagen lattice contraction, probably due to the regulation of fibroblast migration and fibroblast-to-myofibroblast differentiation. Therefore, ILK may be a potential molecular target in regulating wound contraction and treating excessive scar contracture.

Explant culture of sarcoma patients’ tissue

The tissue explant technique here described provides an alternative to enzymatic digestion in order to obtain sarcoma patient-derived cells with a high content of cancer cells in a reasonable time frame. The cells generated by this method can be used for patient-oriented molecular and pharmacological analysis, as well as for the development of new patient-derived animal models.

Apolipoprotein A-1 regulates osteoblast and lipoblast precursor cells in mice

Several lines of evidence propose a link between bone mass and high-density lipoprotein (HDL). The present paper reveals that Apolipoprotein A-1 (ApoA-1), vital component of HDL biosynthesis, serves as a key modulator of osteoblast and lipoblast function and generates changes in the bone cell precursor population that increase adipoblast and decrease osteoblast production, resulting in reduced bone mass and impaired bone quality.

Fibulin-2 is essential for angiotensin II-induced myocardial fibrosis mediated by transforming growth factor (TGF)-β

Fibrosis is an ominous pathological process in failing myocardium. This study demonstrates a regulatory role of fibulin-2 in angiotensin II-induced myocardial fibrosis by enhancing transforming growth factor (TGF)-β activation. Up-regulation of fibulin-2 is likely to interact with latent TGF-β in the extracellular matrix to induce biologically active TGF-β. Fibulin-2 may become a new therapeutic target in preventing heart failure.

Activation of the vitamin D receptor selectively interferes with calcineurin-mediated inflammation: a clinical evaluation in the abdominal aortic aneurysm

This study evaluates the potential of the vitamin D receptor agonist Paricalcitol to quench vascular inflammation. The authors found that a brief course of Paricalcitol has profound effects on local inflammation via reduced T-cell activation and appears to be mediated by an effect on calcineurin-mediated responses.

Perilipin 5 restores the formation of lipid droplets in activated hepatic stellate cells and inhibits their activation

Suppression of perilipin 5 expression is coupled with depletion of lipid droplets (LD) and activation of hepatic stellate cell (HSC) in vitro and in vivo. Introduction of exogenous perilipin 5 inhibits HSC activation by elevating cellular lipid content, restoring LD formation and attenuating oxidative stress. Perilipin 5 might therefore be a candidate for gene therapy in treatment of liver fibrosis.

Modeling fibrosis using fibroblasts isolated from scarred rat vocal folds

In this study, the authors provide detailed, multiplatform characterization of vocal fold (VF) fibroblasts from scarred rat VF mucosae, compared to those from naïve tissue. The scar and naïve VF fibroblast phenotypes are defined and the parameters of an in vitro model for future VF fibrosis research is outlined.
