Genetic inactivation of Cdk7 leads to cell cycle arrest and induces premature aging due to adult stem cell exhaustion

Employing a conditionally inactive gene trap allele, Cdk7's function in regulating cellular proliferation by Cdk1/2-phosphorylation is convincingly dissected from alternative notions on CTD-phosphorylation of RNA Pol II. A new non-catalytic role for ubiquitin ligase RNF8 in unfolding higher-order chromatin structure

RNF8 and RNF168 cooperate during DNA repair to ubiquitylate histone H2A. Strikingly, RNF8 also exhibits a chromatin decondensation activity, which is independent of its ubiquitin-ligase activity and RNF168, but involves a NuRD subunit and which allows assembly of repair factors at DNA lesions.

Differential effects of viral silencing suppressors on siRNA and miRNA loading support the existence of two distinct cellular pools of ARGONAUTE1

Plant viruses encode suppressors (VSRs) that target the host antiviral RNA silencing pathway. Here, VSRs are shown to differentially impair siRNA and miRNA binding to Argonaute1, AGO1, revealing distinct AGO1 pools that are preferentially loaded with either siRNAs or miRNAs.

Co-transcriptional degradation of aberrant pre-mRNA by Xrn2

40S subunit dissociation and proteasome-dependent RNA degradation in nonfunctional 25S rRNA decay

Nonfunctional ribosomes and rRNAs elicit diverse quality control mechanisms. 25S rRNA decay is initiated by the ubiquitination of the nonfunctional 60S subunit and its Cdc48-dependent dissociation from the intact 40S subunit, followed by proteasomal protein degradation and finally 25S rRNA degradation.

Distinct requirement for an intact dimer interface in wild-type, V600E and kinase-dead B-Raf signalling

An intact dimer interface (DIF) is required for both signalling and dimerisation in wild-type Raf, but surprisingly for neither in certain oncogenic Raf versions. The inner membrane histidine kinase EnvZ senses osmolality via helix-coil transitions in the cytoplasm

The bacterial two-component system EnvZ/OmpR governs the osmotic stress response. Increasing osmolality enhances intrahelical H-bonding in the transmembrane kinase EnvZ, leading to helix stabilization, autophosphorylation and activation of downstream signalling.
