Enhancing eNOS activity with simultaneous inhibition of IKKβ restores vascular function in Ins2Akita+/ type-1 diabetic mice

In diabetes, endothelial nitric oxide synthase (eNOS) produces superoxide rather than NO. This paper describes a treatment for enhancing NO availability in the type- 1 diabetic Ins2Akita+/− mouse model. Addition of salsalate, a negative regulator of inhibitor kappa B kinase, to a diet supplemented with L-arginine and sepiapterin, a precursor of the eNOS cofactor tetrahydrobiopterin, significantly improves vasorelaxation of thoracic/abdominal aorta in these mice.

HDAC is essential for epigenetic regulation of Thy-1 gene expression during LPS/TLR4-mediated proliferation of lung fibroblasts

Lipopolysaccharide (LPS)-induced proliferation of lung fibroblasts correlates with loss of expression of thymocyte differentiation antigen-1 (Thy-1), accompanied by deacetylation of histones H3 and H4 at the Thy-1 gene promoter region. This paper shows that LPS down-regulates Thy-1 gene expression by activating histone deacetylases (HDACs) via toll-like receptor 4 (TLR4) signaling, which contributes to Thy-1 gene silencing and lung fibroblast proliferation, leading to pulmonary fibrosis.

Tissue-type plasminogen activator suppresses activated stellate cells through low-density lipoprotein receptor-related protein 1 OPEN

This study shows that in livers subjected to acute carbon tetrachloride poisoning, tissue-type plasminogen activator/lipoprotein receptor-related protein 1 (t-PA/LRP1) signaling through the Akt1 kinase is involved in the regression of activated stellate cells during repair. This finding is unique; previous studies in the kidney show that t-PA/LRP1 signaling through ERK, or extracellular-signal-regulated kinases, is pro-fibrotic.

Pivotal role of liver sinusoidal endothelial cells in NAFLD/NASH progression

This study demonstrates that liver sinusoidal endothelial cell (LSEC) injury plays a “gatekeeper” role in the progression from simple steatosis to the early nonalcoholic steatohepatitis stage. The authors show that LSEC injury appears during the simple steatosis phase, and precedes the appearance of activated Kupffer cells and hepatic stellate cells.

Galectin 3 regulates HCC cell invasion by RhoA and MLCK activation

Galectin-3, a β-galactoside-binding lectin highly expressed in hepatocellular carcinoma (HCC) tissues, regulates HCC invasion and cell motility via mediation of the GTPase RhoA activation and myosin light chain 2 phosphorylation and the consequent reorganization of the actin cytoskeleton. These findings suggest that galectin-3 could be a diagnostic marker and therapeutic target for HCC.

Endoplasmic reticulum stress as a novel target to ameliorate epithelial-to-mesenchymal transition and apoptosis of human peritoneal mesothelial cells

Endoplasmic reticulum (ER) stress induces epithelial-to-mesenchymal transition (EMT) and apoptosis in peritoneal mesothelial cells in a time- and dose-dependent manner. Blocking ER stress ameliorates transforming growth factor-β1-induced EMT and apoptosis, demonstrating the role of ER stress as an adaptive response to peritoneal damage. Therefore, the modulation of ER stress is a novel approach to ameliorate peritoneal fibrosis.

Inhibition of dipeptidyl peptidase IV protects tacrolimus-induced kidney injury

Tacrolimus is a maintenance immunosuppressant that is widely used in renal transplant recipients. Current results reveal that inhibition of dipeptidyl peptidase IV (DPP IV) is effective in improving tacrolimus-induced renal damage. Therefore, a targeted therapy aimed at inhibiting DPP IV may be a useful new approach in the management of both nephropathy and diabetes in renal-transplant patients receiving tacrolimus.

Investigation of the potential of Raman spectroscopy for oral cancer detection in surgical margins

This study shows how well Raman spectroscopy enables discrimination between oral cavity squamous cell carcinoma and surrounding healthy tissue structures. This knowledge supports the development of robust and reliable classification algorithms for future implementation of Raman spectroscopy in clinical practice.

Differential miRNA expressions in peripheral blood mononuclear cells for diagnosis of lung cancer

Because peripheral blood mononucleated cells (PBMCs) act as the first line of defense against malignancy in immune system, their dysfunction may occur as an early event in cancer immunogenicity or immune evasion. This paper shows that differential miRNA expression of PBMCs may provide a new diagnostic approach for the early detection non-small cell lung cancer.

Heterogeneity of subordination of the IL-18/IFN-γ axis to caspase-1 among patients with Crohn’s disease

To determine whether subordination of the Th1 response to the inflammasome pathway is maintained in intestinal inflammation, the authors employed inflamed mucosa explant cultures from patients with Crohn's disease (CD). Th1 cytokine production was assessed in relation with inflammation intensity. Subordination of the Th1 response to caspase-1, effector of the inflammasome, was determined in explant cultures subjected to pharmacological inhibition of caspase-1. This model demonstrated heterogeneity of subordination of the Th1 response to caspase-1 among CD patients, and may be a screening tool for personalized medicine.
