Chronic traumatic encephalopathy: A paradigm in search of evidence?

This editorial review highlights the evolving paradigm surrounding chronic traumatic encephalopathy (CTE), and gaps in knowledge which require further study. An in-depth understanding of CTE by the pathology community is critical to scientific investigation, as current concepts surrounding this entity have broad implications in society, from athletic participation to armed conflict.

Cavin-1 and Caveolin-1 are both required to support cell proliferation, migration and anchorage-independent cell growth in rhabdomyosarcoma

In this paper, the authors employed human rhabdomyosarcoma (RMS) cell lines as well as mouse models that faithfully recapitulate the onset and progression of this soft tissue sarcoma in order to determine mechanisms of tumor growth and invasion. They found that cavin-1, which forms plasma membrane complexes with caveolin-1 to form caveolae, contributes to sustain proliferation, migration and anchorage-independent growth of RMS cells.

RNA sequencing of sarcomas with simple karyotypes: identification and enrichment of fusion transcripts

With the aim of identifying new gene fusions, eight sarcomas with simple karyotypes were subjected to RNA sequencing. By using three different algorithms, three novel and two previously known fusions could be identified. However, only two of the confirmed fusions were reported by more than one algorithm, emphasizing the importance of using multiple tools to analyze RNA sequencing data.

Gammaherpesvirus infection modulates the temporal and spatial expression of SCGB1A1 (CCSP) and BPIFA1 (SPLUNC1) in the respiratory tract OPEN

Scgb1a1 and Bpifa1 encode potential respiratory innate defense proteins. Employing a new, more authentic mouse model of γ-herpesvirus infection, the authors show that the M3 chemokine-binding protein contributes significantly to viral pathogenesis by regulating levels of both SCGB1A1 and BPIFA1. They also reveal that BPIFA1 expression occurs in bronchiolar exocrine cells along with SCGB1A1, but that BPIFA1 is stored within granules.

Decreased proteasomal function accelerates cigarette smoke-induced pulmonary emphysema in mice

This paper reveals that the pathogenesis of chronic obstructive pulmonary disease (COPD) is related to oxidative stress-induced apoptosis in alveolar cells. Transgenic (Tg) mice with decreased proteasomal chymotrypsin-like activity experience accelerated development of cigarette smoke (CS)-induced pulmonary emphysema. Morphologically, CS-exposed Tg mice show remarkable air space enlargement, increased foci of inflammation and apoptotic cells in the alveolar walls of the affected lungs.

High-mobility group box 1 accelerates lipopolysaccharide-induced lung fibroblast proliferation in vitro: involvement of the NF-κB signaling pathway

The mechanism underlying lipopolysaccharide (LPS)-induced aberrant proliferation of lung fibroblasts in Gram-negative bacilli-associated pulmonary fibrosis is examined in this study. The authors show that LPS-induced lung fibroblast secretion of high mobility group box 1 protein augments the pro-proliferative effects of LPS, and may exacerbate pulmonary fibrosis through the activation of the toll-like receptor 4/NF-κβ signaling pathway and its downstream targets.

ORAL AND GASTROINTESTINAL SYSTEMS

Triptolide abrogates growth of colon cancer and induces cell cycle arrest by inhibiting transcriptional activation of E2F

In this paper, the authors demonstrate that minnelide, a water-soluble analog of triptolide, is effective against colon cancer in multiple animal models of colon cancer. Mechanistically, they show that at low concentrations, triptolide induces apoptotic cell death but at higher concentrations it induces G1 cell cycle arrest by inhibiting transcriptional activation of E2F1 through modulation of events downstream of DNA binding.

Osteopontin depletion decreases inflammation and gastric epithelial proliferation during Helicobacter pylori infection in mice

The functional roles of osteopontin (OPN) using a mouse model of Helicobacter pylori (HP)-induced gastritis were investigated in this study. OPN exerts a considerable influence on HP-induced gastritis by contributing the production of cytokines and macrophage infiltration. Moreover, OPN-mediated activation of the MAP kinase pathway in gastric epithelial cells might contribute to epithelial changes following HP infection.

Phenotypic diversity of patient-derived melanoma populations in stem cell medium

Culture of patient-derived melanoma cells in stem cell media has been employed as a method to study melanoma biology, as the properties of parent melanomas are well preserved in patient-derived melanospheres. However, during prolonged culturing, when melanospheres are replaced by cell aggregates, the original heterogeneity is substantially lost. This should be considered when aggregates instead of melanospheres are used to determine melanoma properties and cellular responses to drugs.

Development and characterization of human-induced pluripotent stem cell-derived cholangiocytes

This study demonstrates a novel approach to differentiate induced pluripotent stem cells into cholangiocytes, the epithelial cells that line biliary tree. After extensive characterization of the cells, the study shows that stem cell-derived cholangiocytes can engraft within liver in vivo after retrograde intra-biliary infusion. This work represents a platform for disease modelling, pharmacologic testing, and individualized regenerative therapies for the cholangiopathies.
