Dimethyloxalyglycine stimulates the early stages of gastrointestinal repair processes through VEGF-dependent mechanisms

Dimethyloxalylglycine (DMOG) inhibits prolyl-4-hydroxylase domain enzymes, including hypoxia inducible factor. In vivo and in vitro gut injury models show that DMOG works in a VEGF-dependent manner, stimulating cell migration and proliferation, decreasing indomethacin- and restraint-induced injury. DMOG may therefore be useful in stabilizing or repairing gut mucosa.

FHL2 expression in peritumoural fibroblasts correlates with lymphatic metastasis in sporadic but not in HNPCC-associated colon cancer

Four and a half LIM domain protein-2 expression in myofibroblasts is associated with enhanced migratory capacity and lymphatic metastasis in sporadic colon cancer, but not in peritumoural myofibroblasts of sporadic and hereditary nonpolyposis colorectal cancer. These findings may explain the less invasive and more protruding tumor margin of microsatellite-instable carcinomas.

Norcantharidin inhibits the expression of extracellular matrix and TGF-β1 in HK-2 cells induced by high glucose independent of calcineurin signal pathway

Norcantharidin, an inhibitor of calcineurin, attenuates renal tubulointerstitial fibrosis in rat models of diabetic nephropathy through inhibition of the expression of extracellular matrix and of TGF-β1 in tubules. However, the anti-fibrogenic effect of norcantharidin on tubular interstitium is independent of the calcineurin signaling pathway, so the mechanism is not known.

Netrin-1 overexpression in kidney proximal tubular epithelium ameliorates cisplatin nephrotoxicity

Currently there are no treatments available for cisplatin-induced acute kidney injury. Overexpression of netrin-1, a laminin-related protein, reduces cisplatininduced kidney injury by suppressing inflammation and apoptosis in renal tubular epitheliam both in vivo and in vitro. Therefore, netrin-1 may be a useful therapeutic agent for treating cisplatin nephrotoxicity.

Spleen tyrosine kinase promotes acute neutrophil-mediated glomerular injury via activation of JNK and p38 MAPK in rat nephrotoxic serum nephritis

Spleen tyrosine kinase (Syk) is activated in glomerular neutrophils in human and experimental acute antibody-dependent glomerular injury. Administration of a Syk inhibitor reduces JNK and p38 MAPK signaling, with suppression of glomerular thrombosis and neutrophil recruitment, resulting in abrogation of proteinuria in rat nephrotoxic serum nephritis.

Roles of β-catenin signaling in phenotypic expression and proliferation of articular cartilage superficial zone cells

The mechanisms regulating maintenance of articular cartilage remain unclear, but the superficial zone of articular cartilage is known to be involved. In vivo and in vitro mouse systems are used to demonstrate that β-catenin signaling is required for proliferative activity and differentiation of superficial zone articular cartilage cells.

Erythropoietin improves skin wound healing and activates the TGF-β signaling pathway

Erythropoietin (EPO) accelerates skin wound healing in mice by activating the TGF-β signaling pathway. EPO treatment enhances phosphorylation of Smad2 and Smad3 in fibroblasts and in wound tissue. Blockade of TGF-β attenuates EPO-induced acceleration of wound epithelialization and markedly reverses EPO effects on expression of TGF-β1 and α-smooth muscle actin.

Dual role of the mitochondrial protein frataxin in astrocytic tumors

Although the mitochondrial protein frataxin acts as a tumor suppressor in carcinomas, frataxin expression in glioma cells accelerates tumor growth in nude mice. At the same time, chemosensitivity of glioma cells is increased. Both effects may be the result of changes in reactive oxygen species levels and antioxidative defenses.

A novel origin for granulovacuolar degeneration in aging and Alzheimer's disease: parallels to stress granules

As a marker for stress granules, the phosphorylated ribosomal protein S6 (pS6) localizes to granulovacuolar degeneration in Alzheimer's disease. Further, pS6-positive granules are never associated with apoptotic neurons, and contain less RNA oxidation. These results suggest that granulovacuolar degeneration parallels stress granules and are a neuroprotective response to chronic oxidative stress in Alzheimer's disease brain.

Microfluidics-based capture of human neutrophils for expression analysis in blood and bronchoalveolar lavage

A novel microfluidic cassette procedure for rapid isolation of neutrophils is compared to the traditional laborious method. The new procedure appears equivalent to the standard method of neutrophil isolation for genomic analysis, and identifies differences in gene expression secondary to the source of the cells or in vivo or ex vivo stimuli.

SOX2 is overexpressed in stem-like cells of human lung adenocarcinoma and augments the tumorigenicity

The stem cell transcription factor SOX2 is ectopically overexpressed in lung adenocarcinomas. It is preferentially expressed in cancer-stem cells isolated from lung adenocarcinoma cells, and enhances tumorigenicity of lung cancer cell lines. Therefore, SOX2 may have a role in tumorigenicity of lung adenocarcinoma and is a potential target for treatment.
