The mitochondrial paradigm for cardiovascular disease susceptibility and cellular function: a complementary concept to Mendelian genetics

This review explores the relationship between mitochondrial genetics and function in cardiovascular disease susceptibility with respect to the evolutionary pressures that likely influenced mitochondrial-nuclear relationships. Moving forward, researchers should consider these factors with respect to experimental design and data interpretation when exploring the basis for individual disease susceptibility.

The nuclear receptor CAR modulates alcohol-induced liver injury

The constitutive androstane receptor (CAR) is essential in xenobiotic detoxification. While CAR/ mice are highly susceptible to alcohol-induced liver injury, pre-activation of CAR enhances the hepatic toxicity of alcohol in wild type but not CAR/ mice. Drug-induced CAR activation thus enhances alcohol-induced liver injury by suppressing expression of enzymes that metabolize alcohol.

Inhibition of active autophagy induces apoptosis and increases chemosensitivity in cholangiocarcinoma

Autophagy is activated in cholangiocarcinoma cells that are resistant to chemotherapeutic agents. Inhibition of autophagy suppresses tumor growth, induces apoptosis and sensitizes cholangiocarcinoma cells to chemotherapy-induced cell death. Thus, regulation of autophagy may influence tumor progression and enhance the therapeutic activity of anti-tumor drugs.

Post liver transplantation acute kidney injury in a rat model of syngeneic orthotopic liver transplantation

Acute kidney injury is a frequent complication after liver transplantation. A rat model of syngeneic orthotopic liver transplantation shows that the systemic inflammatory response induced by transplantation is the initiating factor for such kidney damage. Consequently, protection of the liver graft and inhibition of the systemic inflammatory response are vital in reducing the risk of post-liver transplantation acute kidney injury.

Loss of VOPP1 overexpression in squamous carcinoma cells induces apoptosis through oxidative cellular injury

VOPP1 is a recently discovered protein that is over-expressed in a variety of cancers and its over-expression exerts a pro-survival effect . Reducing VOPP1 protein levels in carcinoma-derived cell lines results in oxidative stress, which eventually induces the execution of the apoptotic program.

Nuclear localization of maspin is essential for its inhibition of tumor growth and metastasis

The role of maspin localization in tumor growth and metastasis is revealed by expressing maspin or nuclear-excluded maspin in cells that lack endogenous maspin. Exclusion of maspin from the nucleus abolishes metastasis suppression; only the nuclear form is associated with diminished levels of colony-stimulating factor-1, a cytokine involved in tumor growth and invasion.

Hydrogen sulfide inhibits IL-8 expression in human keratinocytes via MAP kinase signaling

Hydrogen sulfide reduces the basal expression of IL-8, and interferes with IL-17 and IL-22-induced IL-8 production in human keratinocytes. These new observations, taken together with the known anti-inflammatory activity of H2S, are relevant to understanding previously unexplained effects of sulfur-rich balneotherapy in the treatment of psoriasis.

Functional characterization of osteosarcoma cell lines provides representative models to study the human disease

Commonly used osteosarcoma cell lines are characterized functionally and phenotypically in vitro and in vivo to determine their usefulness as model systems for the study of osteosarcoma. Upon subcutaneous xenotransplantation, only a subset of the lines displays characteristics of human osteosarcoma, such as extra cellular matrix production, angiogenesis and metastasis.

Absence of Thy-1 results in TGF-β induced MMP-9 expression and confers a profibrotic phenotype to human lung fibroblasts

Fibroblasts from fibroblastic foci in idiopathic pulmonary fibrosis are Thy-1-negative, while most fibroblasts from normal lungs are Thy-1-positive. Loss of Thy-1 induces a fibrogenic phenotype, including increased proliferative and migratory capacity and contraction of collagen. On a molecular level, the Thy-1-negative cells exhibit TGF-β-induced MMP-9 expression, creating a fibrogenic positive feedback loop.

Loss of functional albumin triggers acceleration of transthyretin amyloid fibril formation in familial amyloidotic polyneuropathy

Transthyretin (TTR)-related familial amyloidotic polyneuropathy is characterized by systemic accumulation of TTR amyloid fibrils. The first evidence is presented that albumin, the most abundant protein in plasma, acts as an antioxidant and plays an inhibitory role in the TTR amyloid formation process.

Bone marrow cell-mediated production of transgenic chickens

The successful production of chickens using cells trans-differentiated from adult chicken bone marrow cells transplanted into the testes is demonstrated. This is a novel approach to the production of transgenic poultry, and is a promising advance in stem cell and developmental biology.

Development of an in vitro model of myotube ischemia

A model for the study of potential therapeutic agents for critical limb ischemia is described. Monolayers of C2C12 myoblasts can be differentiated into mature myotubes and used to simulate parameters that occur in skeletal muscle ischemia. As proof-of-concept, erythropoietin exhibits tissue protective effects in the model.

Quantitative assessment shows loss of antigenic epitopes as a function of pre-analytic variables

Quantitative assessment of the effect of delayed formalin fixation on surgical resections, using biopsies as controls, suggests that measurement of phosphoproteins by immunological methods is dramatically affected by pre-analytic variables. Core needle biopsies suffer less pre-analytic variation and appear to be more valuable for assessment of biomarkers.
