Role of β4 integrin phosphorylation in human invasive squamous cell carcinoma: regulation of hemidesmosome stability modulates cell migration

Hemidesmosomes anchor epithelial cells to the basement membrane and are disassembled during cell migration and carcinoma invasion. The increased phosphorylation of β4 integrin in squamous cell carcinoma potentially plays an important role in hemidesmosome disassembly, allowing the cells to migrate and disseminate.

Nodular fasciitis: a novel model of transient neoplasia induced by MYH9-USP6 gene fusion

Nodular fasciitis is a self-limited myofibroblastic proliferation historically viewed as a reactive process of uncertain etiology. Most cases of nodular fasciitis are now shown to harbor the fusion gene MYH9-USP6, making it the first example of a selflimited human disease characterized by a recurrent somatic gene fusion event.

Cell-specific effects of TNF-α and IL-1β on alkaline phosphatase: implication for syndesmophyte formation and vascular calcification

TNF-α and IL-1β stimulate tissue non-specific alkaline phosphatase activity in vascular smooth muscle cells through PPARc inhibition. In contrast, inhibition of PPARc by both cytokines decreases the enzyme's activity in entheseal chondrocytes from organotypic cultures of mouse bones, suggesting that in ankylosing spondylitis, TNF-α and IL-1β inhibit syndesmophyte formation.

Expression of Snail and Slug in renal cell carcinoma: E-cadherin repressor Snail is associated with cancer invasion and prognosis

In renal cell carcinoma, Snail protein expression is associated with tumor stage and poor prognosis. Targeting Snail expression in the 786-O RCC cell line with siRNA inhibits invasion in vitro, which suggests that Snail plays an important role in invasion and metastasis, and that gene silencing is a potential therapeutic strategy.

Inhibition of inflammation by pentosan polysulfate impedes the development and progression of severe diabetic nephropathy in aging C57B6 mice

Pentosan polysulfate (PPS) treatment prevents the progression of nephropathy in streptozotocin-induced diabetes in aging C57B6 mice by decreasing albuminuria, renal macrophage infiltration and TNF-α expression. These effects are associated with improvement of histopathologic changes and renal function. PPS suppresses inflammation mediated by TNFa, high glucose, and advanced glycation end products.

MicroRNA-148a is down-regulated in human pancreatic ductal adenocarcinomas and regulates cell survival by targeting CDC25B

miRNA-148a is significantly down-regulated in human pancreatic ductal adenocarcinomas, and decreased expression leads to a higher proliferation rate and increased colony formation. The dual specificity protein phosphatase CDC25B is verified as a miR-148a target, indicating that this miRNA exhibits tumor-suppressor function in pancreatic ductal adenocarcinomas.

Development and characterization of xenograft model systems for adenoid cystic carcinoma

The study of adenoid cystic carcinoma (ACC) has been hampered by the lack of model systems. The development and characterization of 12 distinct xenograft lines of ACC are outlined. The models recapitulate the morphologic, gene expression and genetic characteristics of primary ACC tumors, providing the basis for further discovery and hypothesis-driven research in this disease.

High-throughput detection of fusion genes in cancer using the Sequenom MassARRAY platform

Identification of recurrent expressed fusion genes through FISH and RT-PCR are time-consuming and prone to artefacts. A novel high-throughput strategy for the detection of recurrent fusion genes based on the Sequenom MassARRAY platform is described, and shows promise as a high-throughput screening tool for these genes in human cancer.

Induction of metastatic cancer stem cells from the NK/LAK-resistant floating, but not adherent, subset of the UP-LN1 carcinoma cell line by IFN-γ

Floating and adherent subsets of a lymph node metastatic carcinoma cell line displayed CD44high/CD24low and CD44low/CD24high phenotypes, which respectively corresponded to monodispersed and compact tumor cells in the patient's metastatic lesion. The floating subset is the major niche for cancer stem cells, and upon IFN-γ treatment could be induced to CXCR4+ metastatic cells.

Elf3 plays a role in regulating bronchiolar epithelial repair kinetics following Clara cell-specific injury

Elf3 is an epithelial-specific transcription factor that is highly expressed during fetal lung development. The kinetics of cell proliferation and Clara cell renewal are delayed within the regenerating bronchiolar airway epithelium of Elf3-deficient mice after injury, which reveals an important role for Elf3 in the regulation of epithelial repair.

Neonatal chlamydial pneumonia induces altered respiratory structure and function lasting into adult life

Neonatal pulmonary Chlamydia muridarum infection in mice leads to reduced numbers of alveoli and impaired pulmonary function as adults. Antimicrobial therapy of infected neonates partially alleviates the Chlamydia-induced pulmonary dysfunction, supporting the role of Chlamydia in pathological sequelae following respiratory infection.

Pathogenic role of B cells in the development of diffuse alveolar hemorrhage induced by pristane

A single intraperitoneal injection of pristane induces alveolar hemorrhage in C57BL/6 mice within two weeks. The effect is independent of MyD88, TLR7, FcγR, TCR, and Fas; however, absence of B-cells reduces the prevalence. This model can be useful for determining the pathogenesis of this life-threatening complication and for testing new therapies.
